The major purpose of this article is to evaluate oligochitosan coated cerium oxide nanoparticles (OCCNPs) alginate laden injectable hydrogels and their potential treatment for age-related macular degeneration (AMD). The water soluble OCCNPs were loaded within injectable hydrogels as antioxidative agents. The release of OCCNPs from hydrogel, radical scavenging properties, and biocompatibility were evaluated and calculated in vitro. The effects of OCCNP laden hydrogel downregulating expression of angiogenic proteins and proinflammatory cytokines were quantified in human retinal pigment epithlium-19 (ARPE-19) and umbilical endothelium cell lines...

Idiopathic pulmonary fibrosis (IPF) is a devastating disease and the pathogenesis of IPF remains unclear. Our previous study indicated that miR-5100 promotes the proliferation and metastasis of lung epithelial cells. In this study, we investigated the effect and mechanism of miR-5100 on bleomycin (BLM)-induced mouse lung fibrosis and transforming growth factor β (TGF-β1) or epidermal growth factor (EGF) induced EMT-model in A549 and Beas-2B cells. The elevated level of miR-5100 was observed in both the mouse lung fibrosis tissues and EMT cell model...

BACKGROUND: Human Cytomegalovirus (HCMV) infections can be found throughout the body, especially in epithelial tissue. Animal model was established by inoculation of HCMV (strain AD-169) or coinoculation with Hepatitis E virus (HEV) into the ligated sacculus rotundus and vermiform appendix in living rabbits. The specimens were collected from animals sacrificed 1 and a half hours after infection. RESULTS: The virus was found to be capable of reproducing in these specimens through RT-PCR and Western-blot...

Idiopathic pulmonary fibrosis (IPF) is a progressive fibrotic lung disease with high mortality. Active TGFβ1 is considered central to the pathogenesis of IPF. A major mechanism of TGFβ1 activation in the lung involves the epithelially restricted αvβ6 integrin. Expression of the αvβ6 integrin is dramatically increased in IPF. How αvβ6 integrin expression is regulated in the pulmonary epithelium is unknown. Here we identify a region in the β6 subunit gene (ITGB6) promoter acting to markedly repress basal gene transcription, which responds to both the Ets domain-containing protein Elk1 (Elk1) and the glucocorticoid receptor (GR)...

Mutations in the photoreceptor tetraspanin gene peripherin-2/retinal degeneration slow (PRPH2/RDS) cause both rod- and cone-dominant diseases. While rod-dominant diseases, such as autosomal dominant retinitis pigmentosa, are thought to arise due to haploinsufficiency caused by loss-of-function mutations, the mechanisms underlying PRPH2-associated cone-dominant diseases are unclear. Here we took advantage of a transgenic mouse line expressing an RDS mutant (R172W) known to cause macular degeneration (MD) in humans...

After presenting both the prevalence and the staging of the age-related macular degeneration (AMD), the disease progression is discussed; it is determined primarily by genetic factors but is also influenced by environmental, demographic and behavioral factors. The genes working by means of the high-density lipoprotein cholesterol (HDL) pathway have critical roles in the drusen initiation in the early AMD stages. After the drusen have accumulated between retinal pigment epithlium (RPE) and Bruch's membrane, the genes acting in the alternative complement pathway are activated so influencing in this way the progression from the intermediate and large drusen to the advanced AMD stages (e...

OBJECTIVE: To observe the expression of MMP-9, TNF-alpha in cholesteatoma and to determine their roles in the destruction of bone. METHOD: Immunohistochemical method (SP) and the computer image quantitative analysis were used to examine the expression of TNF-alpha, MMP-9 in 22 samples from cholesteatoma and 20 normal cases. At the same time, the correlation between the expressions of the two indexes and the erosion of ossicles were observed. RESULT: MMP-9 protein was only expressed in cholesteatoma stromal cells...

Calcium was localized by in situ precipitation with potassium antimonate during epididymal maturation of the mouse and guinea pig sperms. In caput epididymis the calcium in mouse sperm head was mainly localized on the inner surface of tha outer acrosomal membrane (OAM) in preacrosomal region. During the passage from the caput epididymis to the cauda epididymis the calcium amount of mouse sperm did not undergo apparent changes. In comparison with mouse sperm, there were a few fine calcium deposite granules on the inner surface of the guinea pig sperm OAM on the abdomen side at the caput epididymis stage, but these granules disappeared at the cauda epididymis stage...

COMPARATIVE STUDY

1. Endothelin (1 nM-0.3 microM) produced a concentration-dependent contraction of guinea-pig epithelium-containing (intact) trachea (EC50 = 30.9 nM). Endothelin was a less potent agonist than leukotriene D4 (LTD4; EC50 = 0.77 nM), but was more potent than carbachol (EC50 = 0.15 microM) or substance P (EC50 = 1.4 microM). Endothelin was a more potent contractile agent in rat endothelium-denuded aorta (EC50 = 2.1 nM) than in guinea-pig trachea. 2. Endothelin-induced contraction in guinea-pig trachea was unaffected by mepyramine (10 microM), atropine (1 microM), SK&F 104353 (10 microM), a leukotriene receptor antagonist, or SQ 29,548 (1 microM), a thromboxane receptor antagonist...

I used seven eyes obtained postmortem to study the mechanism of fixation of a Binkhorst two-loop iridocapsular lens. During the first postoperative weeks, vitreous pressure and traction on the posterior lens capsule exerted by the zonular fibers pushed the loops of the iridocapsular lens against the iris. The zonular traction maintained the posterior capsule taut so that the vitreous pressure was distributed equally over the entire surface of the capsular diaphragm. Lens epithelial cells proliferated around the margin of the capsular breaks and defects and formed plaques or masses of spindle-shaped cells...

In the animals subjected to the operation but without cortisone injection (Fig. 1, a and Fig. 2, delta), a high proliferative activity in the skin epithelial cells surrounding the autograft was observed during the whole course of the experiment. In the cells of the autograft, the mitotic activity, after inhibition, restored quickly up to the original level, and in 10 days it sharply increased and did not differ from that in the epithelium of the surrounding skin...

In pursuit of a model system in which to determine whether exposure to progesterone is necessary for mammary epithlial cells to develop their differentiative potential, hormone-dependent growth of the mammary epithelial rudiment in adult male mice has been reexplored. The formation of ductal cells can be effected by administration of estradiol in the absence of endogenous progesterone and glucocorticoid, using adrenalectomized-castrated animals. The resulting epithlium contains three times more lactose synthetase activity, per epithelial cell, than that in midpregnant mice...
