A total of 329 novel adamantane derivatives have been synthesized and pharmacologically studied. Among them, the compounds containing halogen-containing aromatic radicals in the second position show the most pronounced effect on animal resistance to the emergencies induced by its habitat and performance. N-(2-adamantyl)-N-(para-bromphenyl)amine (bromantane) possesses a low toxicity, a high ability to enhance the physical and operant working maintenance of animals, to accelerate its recovery in developed fatigue, in hyperthermia and hypoxia in particular...

The positive effect of bromantan (0.5-50 mg/kg) on the efficiency of physical work of mice and rats (swimming test, running on a treadmill) exceeds that of phenamine administered in optimal doses by 1.3-1.6 times. Such an effect of bromantan lasts no less than 24 h. The drug delays decrease of physical efficiency and restores it sooner in repeated extreme running loads, prevents in rats the corresponding electron-microscopic changes in the cardiomyocyte and myocyte mitochondria of the skeletal muscles. The data obtained suggest that the positive effect of bromantan on the physical efficiency is associated not only with its psychostimulating action but also with the membrane-protecting effect...

The action of the new stimulant bromantane on spectra power EEG on Fourier of sensorimotor cortex, dorsal hippocamp and lateral hypothalamus of left and right hemispheres of brain of rat in free behavior was investigated. Bromantane leads to decreases in the total and absolute powers of all frequency bands of EEG spectra, changes structural spectra in the cortex and in hippocamp--decreases the relative power of theta-band and increases the relative power of beta 1, 2-activity. The basic feature of bromantane's action is a two-phase effect (its maximum occurs 2-3 and 6-7 hours after administration), which remains up to 8 h of EEG recording...

The thermoprotective properties of bromantane were studied during overheating. Simultaneously with the known effects on heat exchange and sensitivity to high temperatures bromantane caused a decrease in ranges of heat tolerance, increased the rate of SVTK growth as well as altered protein metabolism and antioxidant protection and also affected the autonomic regulation of heat exchange which were essential in adaptation. At the same time, during ergothermal loading accompanied by any troubles in evaporation and heat exchange, marked thermoprotective efficiency of bromantane was compared for its ability to improve muscle dynamometry and hemodynamics as compared with control values...

The effect of acute and chronic administration (50 mg/kg, p.o.) of a new immunostimulator, bromantan exhibiting psychostimulant features on the content of NE, DA and 5-HT, and their metabolites are studied. Bromantan induced a significant increase in the 5-HT and 5-HIAA content in the frontal cortex and delayed an increase in their content in subcortical brain regions. A stable decrease in the 5-HT and 5-HIAA levels in the cerebellum is observed. The drug also affected the DA parameters of the brain thus suggesting an important role of dopaminergic system in the mechanism of pharmacological effects of the drug...
