Transient pancytopenia as an adverse hematologic reaction due to ticlopidine. We present a case of transient pancytopenia due to ticlopidine in 66-year-old woman who was administrated with ticlopidine as a primary prevention of heart failure. The first sign was a skin rush which was followed by ticlopidine cessation. Two days later she developed septic shock, pneumonia and neutropenia (600 cells/mm3) with decrease of platelats, erytrocytes count and hemoglobin. Blood transfusions were not required. After treatment with antibiotics according to antibiogram the patient recovered and was dismissed after 38 days with normal blood morphology...

Modification of tyrosine residues in extracellular proteins by a sulfate moity plays an important role in many ligand/receptors interactions. In the present work, we describe a unique human monoclonal antibody, termed Y1-scFv, that is specific for a sulfated epitope in the platelat receptor GPIb. The Y1-scFv single chain antibody (scFv) competes with von Willebrand factor (vWF) for binding to human platelets and thus effectively inhibits platelet aggregation. Limited proteolysis of GPIb molecule, using the endoproteases, mocarhagin and cathepsin G, revealed that a seven amino-acid epitope, Tyr-276 to Glu-282, contains the recognition site for Y1-scFv...

The effects of bile from rabbits and deoxycholate on platelat aggregation and platelet morphology were studied. (1) Bile diluted to 1.25%, as well as 0.1% deoxycholate, significantly inhibited ADP-induced aggregation (p less than 0.01). (2) Ring-shaped platelets were observed in preparations treated with bile containing high concentrations of deoxycholate. These platelets lacked the center portion, but maintained the circumferential band of microtubules as revealed by scanning and transmission electron microscopy...
