The administration of neurotoxin thiolactone homocysteine in rats with cobalt-induced epileptogenic focus in the sensomotor cortical region led to the development of secondary generalized convulsions and epileptic state. The pattern was analogous to the manifestation of convulsions in epileptic state in humans. Fenitoin (50 mg/kg) inhibited the development of convulsions, arrested the epileptic state, reduced the number and duration of the secondary generalized tonic--clonic attacks, decreased the behavioral manifestations (focal convulsions, lateral position etc...

We present a 41 woman who underwent an allogeneic bone marrow transplantation as a treatment for a multiple myeloma. After presenting graft-versus-host disease, hyperbilirubinemia, cyclosporine toxic levels, renal failure and peripheral schistocytosis, she developed complex partial seizures and bilateral hypodensity lesions in occipital lobes. Fulminant microangiopathy was diagnosed; though promptly treated with immunoglobulins and fenitoin, she died in few days. Necropsy showed diffuse thrombotic disease and a haematoma in left occipital lobe...

During pregnancy, there are several physiological changes that influence the kinetics of medication administrated during this time. These physiological changes are not reestablished immediately during delivery; so, its concentration in biologic fluids is different when is administered immediately after delivery, than several weeks after. The purpose in this work was to identify the changes of pharmacocinetics constants for fenitoin and carbamacepine, in epileptic patients. When the same dose is maintained, during different times of postpartum...

The authors investigated the effects of the inductors (griseofulvin and fenitoin) and the inhibitors of the cytochrome P-450 (metronidazole, chloramphenicol and phenylbutazone), the effects of the drugs with the purine structure (aminophylline, xanthinol-nicotinate, pentoxiphylline, azathioprine, thioguanine, 6-mercaptopurine), and the effects of several pesticides (lindane, binapicrile, parathion) on some biochemical parameters of the rat liver. The following parameters were determined: viability of hepatocytes, the content of glutathione and cytochrome P-450, and the activity of xanthinoxidase and lipoperoxidase...
