Two compounds, 3'-deoxyinosine and cordycepin, were isolated from Bombyx mori inoculated with Cordyceps militaris. In the bioassay examining cytotoxicity against cancer cells, both compounds showed toxicity against A549, PANC-1, and MCF-7 cancer cells.

Cryptochromes (CRYs) belong to the photolyase/cryptochrome flavoprotein family, which is widely distributed in all kingdoms. A phylogenetic analysis indicated that three Cordyceps militaris proteins [i.e., cryptochrome DASH (CmCRY-DASH), (6-4) photolyase, and cyclobutane pyrimidine dimer (CPD) class I photolyase] belong to separate fungal photolyase/cryptochrome subfamilies. CmCRY-DASH consists of DNA photolyase and flavin adenine dinucleotide-binding domains, with RGG repeats in a C-terminal extension. In this study, a degenerated strain of C. militaris was obtained by subculturing a wild-type strain through 10 successive subcultures. Cordyceps militaris has commercially been cultivated, but its degenerated subcultures have gradually resulted in the reduced production. In this study, the biological characteristics and DNA change of degenerated strains of C. militaris were analyzed in detail. The results showed that the degenerated strains exhibited the lower growth rate, and the deficiency in fruit body formation and pigment production. The degradation of strains was not attributable to DNA changes identified by RAPD and SRAP.
