BACKGROUND: It is important to minimize placebo rates in randomised controlled trials (RCTs) to efficiently detect treatment differences between interventions. Historically, high placebo rates have been observed in clinical trials of ulcerative colitis (UC). A better understanding of factors influencing placebo rates may lead to more informed clinical trial design. OBJECTIVES: A systematic review and meta-analysis was conducted to evaluate placebo response and remission rates in RCTs evaluating UC treatments in adult patients...

Patients with relapsed aggressive non-Hodgkin lymphoma (NHL) are often treated with platinum-based chemoimmunotherapy regimens in preparation for autologous stem cell transplant. We sought to reduce toxicity and maintain efficacy by using oxaliplatin with rituximab, cytarabine and dexamethasone (ROAD) in a phase II clinical trial in patients who had relapsed after one prior regimen. ROAD was delivered q21 days and consisted of rituximab 375 mg/m(2) IV weekly x 4 doses (cycle 1 only); dexamethasone 40 mg PO/IV d2 - 5; oxaliplatin 130 mg/m(2) IV day 2; cytarabine 2000 mg/m(2) IV × two doses on days 2 to 3; and pegfilgrastim 6 mg SC on day 4...

Peer-reviewed journals and conference proceedings publish hundreds of papers that describe new medical imaging algorithms, including, for example, techniques for computer-aided diagnosis or characterization, segmentation, image registration, image reconstruction, and radiomics. It is difficult, if not impossible, to fairly compare the performance of these algorithms as investigators must either use different data sets, or if using the same data, use different implementations of competing algorithms. Grand Challenges facilitate the fair comparison of algorithms by providing a common data set to all participants and by having each participant be responsible for implementation of their own algorithm...
