To make a clear understanding of the in vivo metabolism of propyl gallate in rats and determine the original compounds of the metabolites of Paeoniae Radix Rubra (PRR), the urine and plasma of the rats administrated with propyl gallate were collected, and then the HPLC-DAD-ESI-IT-TOF-MS(n) technique was applied to screen and identify the metabolites of propyl gallate from these bio-samples. In order to provide an experimental foundation for clinical study, we made a preclinical pharmacological investigation of 99mTc-Q3, a novel myocardial perfusion imaging agent, technetium-99m-N,N'-ethylenebis (acetylace-toneiminato) bis (tris (3-methoxy-1-proply) phosphine). After the preparation of this compound, the kinetics of blood clearance in rabbits, biodistribution in mice, measurement of plasma protein binding rate, and myocardial perfusion imaging in dog were carried out. Amphetamine and five N-alkylated homologues were readily metabolized by Mycobacterium smegmatis and the products obtained were identified by gas-liquid chromatography and mass spectrometry. The N-aklylsubstituent had a considerable influence on the degree and mechanism of biotransformation. With the exception of the N-isopropyl derivative, all of the N-alkylated homologues were deaklylated to amphetamine which was then conjugated to the N-acetyl derivative. Pancreases from Syrian golden hamsters treated with N-nitrosobis(2-hydroxy-proply)amine for 10 to 25 weeks were examined under scanning electron microscopy (SEM). Findings indicate that the neoplasms originated from the ductal epithelium and developed progressively. Adenomas were lined by epithelium of differing cells types, ranging from a flat singly ciliated form to cuboidal-columnar types, or to mixed cell populations.
