BACKGROUND: Breast cancer is a complex heterogeneous disease where many distinct subtypes are found. Younger African American (AA) women often present themselves with aggressive form of breast cancer with unique biology which is very difficult to treat. Better understanding the biology of AA breast tumors could lead to development of effective treatment strategies. Our previous studies indicate that AA but not Caucasian (CA) triple negative (TN) breast cancer cells were sensitive to nitrosative stress-induced cell death...

Gliomas are typically characterized by their infiltrative nature, and the prognosis can be linked to the invasive nature of the tumoral cells. Glioblastoma multiforme are very invasive cancers and this contributes to their lethality. The invadopodia are considered essential for their motility. Human glioma cell invadopodia were examined with transmission electron and immunofluorescent microscopy. By electron microscopy, in situ gliomas (fibrillary astrocytoma, anaplastic astrocytoma, glioblastoma multiforme, pilocytic astrocytoma) show mitochondria with a dense matrix condensed configuration, indicating an active state...

Ubiquinone or Co Q(10) is essentially a vitamin like substance and is a cofactor of an enzyme. It is an integral part of the memberanes of mitocondria where it is involved in the energy production. It is a nutrient necessary for the function of every cell of the body especially vital organs of the body like heart, liver, brain etc. Studies have shown that coenzyme Q(10) alters the natural history of cardiovascular illness and has the potential of prevention of cardiovascular diseases through the inhibition of LDL cholesterol oxidation by maintenance of optimal cellular and mitochondrial function throughout the ravages of time internal and external stress...

Mitochondria possess a sophisticated array of Ca(2+) transport systems reflecting their key role in physiological Ca(2+) homeostasis. With the exception of most yeast strains, energized organelles are endowed with a very fast and efficient mechanism for Ca(2+) uptake, the ruthenium red (RR)-sensitive mitochondrial Ca(2+) uniporter (MCU); and one main mechanism for Ca(2+) release, the RR-insensitive 3Na(+)-Ca(2+) antiporter. An additional mechanism for Ca(2+) release is provided by a Na(+) and RR-insensitive release mechanism, the putative 3H(+)-Ca(2+) antiporter...

The freshwater Amphipod Gammarus pulex is a sensitive indicator organism for environmental contamination. In this study, effects of heavy metal lead acetate were investigated by using electron microscopy techniques. Control group was not expose to lead acetate but experimental group was for 96 h. The ultrastructural changes of hepatopancreatic ceca cells were studied in these two groups by electron microscopy techniques analysing cellular structure, structure of organelles and vacuolization. The number of cells based on the toxic effects of lead acetate was increased compared with the control group...

The euglenoids are unicellular eukaryotic flagellates living in a diversity of soils and aquatic environments and ecosystems. This study describes the ultrastructure of an euglenoid isolated from the surface of a boiling mud pool with temperatures ranging from 38 to 98 degrees C and pH 2 - 4. The hot mud pool is located in Area de Pailas de Barro, Las Pailas, Rincón de la Vieja Volcano, Guanacaste, Costa Rica. The morphological characterization of the Euglena pailasensis was performed by SEM and TEM. It was determined that, although the euglenoid was obtained from an extreme volcanic environment, the general morphology corresponds to that of a typical member of Euglena of 30-45 microm long and 8-10 microm wide, with membrane, pellicle, chloroplasts, mitochondria, nucleus, pigments and other cytoplasmic organelles...

Seals are unique among mammals in that newborns have a large pineal gland and extremely high plasma levels of melatonin at birth. Melatonin levels are also high in the seal fetus but decline rapidly during the first few days of life. The aim of the present study was to provide quantitative information about the ultrastructure of the seal pineal gland using fetal, newborn, and adult hooded seals (Cystophora cristata), and newborn and adult harp seals (Phoca groenlandica). The relative and absolute volumes of pinealocytes (Pi), arteries and veins, nerves, connective tissue, capillaries and glial cells, as well as mitocondria and lipid droplets in Pi, were calculated by use of point count analysis...

The aim of this study was to identify the significant sites of action of cadmium on oxidative phosphorylation in potato tuber mitocondria. We simplified the system to three convenient subsystems linked via the production or consumption of a common intermediate, namely protonmotive force. The three subsystems were substrate oxidation, which produces protonmotive force, and the proton leak reactions and the phosphorylation reactions, which consume protonmotive force. By measuring the effect of cadmium on the kinetic response of each subsystem to protonmotive force (top-down elasticity analysis), we found that cadmium stimulated proton leak reactions and strongly inhibited substrate oxidation, but had no measurable effect on the phosphorylation reactions...

A non-enzymatic method is presented for isolating mitochondria from small-sized human cardiac samples, including ventricular needle biopsies of 15-25 mg of wet weight. Electron microscopy demonstrates that these fractions are rich in structurally well preserved mitochondria. Calcium and magnesium levels of fractions are determined by atomic absorption flame spectroscopy. Comparative analyses are made in similar fractions of the mouse ventricle. Calcium concentrations of mitocondria isolated in the presence of ruthenium red do not differ significantly between the human auricle and ventricle, averaging 61 nmol Ca/mg protein and 68 nmol Ca/mg protein, respectively...

We examined the cerebellum of the rats being the experimental hypothyroidism (cretinism) on the 20th day, 35th day and 60th day by the light and electron microscope. The remarkable findings were observed to the experimental group on the 20th day. The maturational states of this group correspond to that of the 16th day of the control group. On the other hand, the remarkable pathological findings were not observed on the 35th and 60th day of the experimental group. According to the findings of the experimental rats on the 20th day, the changes of the cells were the retention of the external granule cells and the maturational changes of the internal granule cells...

This paper presents a study of the ultrastructure of the epithelium of the soft plate of the rat. General morphology, ratio and localization of ribosomes, filaments, mitocondria, spaces of stratum basale, stratum spinosum and stratum granulosum, are described. The stratum corneum presents modified desmosomes, no organelles and enlarged intercellular spaces. Cornified cells are constituted of a filament-like material, densely packed in an amorphous matrix.

The authors, in this second part of the oxygen story, resolutely cross the borders of the biophysical field, and face the origins and becoming of life, the stages of which are synthesized in "casket" terms, unusual for surgeons: "protobionts", "procariots", "cyanobacteria", "chlorophyll", "caroteonides", "fermentation", "anaerobic glycolysis", "eucariots", "respiratory chain", "mitocondria". This is not an unconventional biological exercise, but the effort to give clinics a more legible ground, a sort of common denominator of the most different pathologies, and, among these ones, at the first place, just those of the specialistic branch, also less frequenter of biology, that is, surgery...

The origin of peroxisomes in yeast organisms is still unknown. These organelles are believed to be formed, similar to animal cells, from the endoplasmatic reticulum. However, this has not been confirmed directly. Peroxisomes are often found to be in contact with channels of the endoplasmatic reticulum and, in our experiments, with mitochondria of yeast organisms, especially those which utilize oleic acid, n-alkanes and methanol as a sole source of carbon. In Rhodotorula, peroxisomes are characterized by the same "bean" configuration and paired arrangement imitating "copulation" as mitocondria...

The effect of amphotericin B and nystatin on mitochondria isolated from the dog kidneys was studied. It was shown that incubation of the isolated kidney mitocondria with amphotericin B or nystatin at a wide range of their concentrations, i.e. from 0.1 to 100 gamma per 0.5 mg of the mitochondrial protein did not affect the activity of succinate dehydrogenase of these cell microstructures. The absence of changes in the succinate dehydrogenase activity of the mitochondria under the effect of the polyenic antibiotics is significant from the viewpoint of elucidating their molecular mechanisms of the nephrotoxic effect...
