Multiple mechanisms for the gut microbiome contributing to the pathogenesis of nonalcoholic fatty liver disease (NAFLD) have been implicated. Here, we aim to investigate the contribution and potential application for altered bile acids (BA) metabolizing microbes in NAFLD by post hoc analysis of whole metagenome sequencing (WMS) data. The discovery cohort consisted of 86 well-characterized patients with biopsy-proven NAFLD and 38 healthy controls. Assembly-based analysis was performed to identify BA-metabolizing microbes...

Bile acids are detergent molecules that solubilize dietary lipids and lipid-soluble vitamins. Humans synthesize bile acids with α-orientation hydroxyl groups which can be biotransformed by gut microbiota to toxic, hydrophobic bile acids, such as deoxycholic acid (DCA). Gut microbiota can also convert hydroxyl groups from the α-orientation through an oxo-intermediate to the β-orientation, resulting in more hydrophilic, less toxic bile acids. This interconversion is catalyzed by regio- (C-3 vs. C-7) and stereospecific (α vs...

7α-Hydroxysteroid dehydrogenase (7α-HSDH) plays an important role in the efficient biotransformation of taurochenodeoxycholic acid (TCDCA) to tauroursodeoxycholic acid (TUDCA). In this paper, a novel NADP(H)-dependent 7α-HSDH (named J-1-1) was discovered, heterologously expressed in Escherichia coli and biochemically characterized. J-1-1 exhibited high enzymatic activities. The specific activities of J-1-1 toward TCDCA, glycochenodeoxycholic acid (GCDCA) and ethyl benzoylacetate (EBA) were 188.3 ± 0.2, 217...

7α-Hydroxysteroid dehydrogenase (7α-HSDH) is one of the key enzymes in the catalytic reaction of taurochenodeoxycholic acid (TCDCA). To improve the activity of immobilized 7α-HSDH, the microenvironment of immobilized 7α-HSDH was modified with epoxy resin and ethanediamine (EDA). The amino-epoxy support was characterized by Fourier transform infrared (FTIR), Spectrometer elemental analysis (EA), scanning electron microscopy (SEM), contact angle (CA), and Zetasizer. The effects of the immobilization of 7α-HSDH on the amino-epoxy resin and epoxy resin were studied...

The present study was undertaken to investigate the toxic effect of sodium fluoride (NaF)- and fluoride (F)-contaminated groundwater on male reproduction and it's reversibility in male rats. Adult male rats were orally treated with different concentrations of NaF- (1 mg, 5 mg, and 10 mg/kg/bw/rat) and F-contaminated groundwater for 52 days and after the confirmation of F-induced damage, the rats were allowed for recovery studies for 52 days. Exposure of NaF- and F-contaminated groundwater caused significant decline in total sperm count, sperm motility, serum concentration of testosterone, activities of testicular 3β-HSDH, counts of type A spermatogonia, preleptotene spermatocytes, midpachytene spermatocytes, elongated spermatids and round spermatids, activities of testicular and spermatozoa SOD and CAT, and increase in sperm abnormality and concentration of MDA of testis and spermatozoa compared to controls...

Heat and calcium treatments are used to improve postharvest fruit longevity; however, the exact mechanism remains poorly understood. Anaerobic bacteria inhabiting the human gastrointestinal tract have evolved various enzymes that modify host-derived steroids. The bacterial steroid-17,20-desmolase pathway cleaves the cortisol side-chain, forming pro-androgens predicted to impact host physiology. Bacterial 20β-hydroxysteroid dehydrogenase (20β-HSDH) regulates cortisol side-chain cleavage by reducing the C-20 carboxyl group on cortisol, yielding 20β-dihydrocortisol. Recently, the gene encoding 20β-HSDH in Butyricicoccus desmolans ATCC 43058 was reported, and a non-redundant protein search yielded a candidate 20β-HSDH gene in Bifidobacterium adolescentis strain L2-32...

Strains of Eggerthella lenta are capable of oxidation-reduction reactions capable of oxidizing and epimerizing bile acid hydroxyl groups. Several genes encoding these enzymes, known as hydroxysteroid dehydrogenases (HSDH) have yet to be identified. It is also uncertain whether the products of E. lenta bile acid metabolism are further metabolized by other members of the gut microbiota. We characterized a novel human fecal isolate identified as E. lenta strain C592. Bile acids are important cholesterol-derived nutrient signaling hormones, synthesized in the liver, that act as detergents to solubilize dietary lipids. Bile acid 7α-dehydroxylating gut bacteria generate the toxic bile acids deoxycholic acid and lithocholic acid from host bile acids. The ability of these bacteria to remove the 7-hydroxyl group is partially dependent on 7α-hydroxysteroid dehydrogenase (HSDH) activity, which reduces 7-oxo-bile acids generated by other gut bacteria. 3α-HSDH has an important enzymatic activity in the bile acid 7α-dehydroxylation pathway...

BACKGROUND: Enhancing thermostability of the 7α-Hydroxysteroid dehydrogenases (7α-HSDHs) is beneficial to its industrial application broadly. For protein engineering to enhance thermostability the nonrational strategy, directed evolution, has been applied in obtaining more stable proteins through error-prone PCR or DNA rearrangement generating random mutations. However, the successful application of directed evolution needs to build a large mutant library. Site-directed mutations of CA 7α-HSDH had been performed to probe the relationship between the compactness increasing and thermostability enhancing...
