Quantifying metal and nanoparticle (NP) biouptake and distribution on an individual cellular basis has previously been impossible, given available techniques which provide qualitative data that are laborious to acquire and prone to artifacts. Quantifying metal and metal NP uptake and loss processes in environmental organisms will lead to mechanistic understanding of biouptake and improved understanding of potential hazards and risks of metals and NPs. In this work, we present a new technique, single cell inductively coupled plasma mass spectrometry (SC-ICP-MS), which allows quantification of metal concentrations on an individual cell basis down to the attogram (ag) per cell level...

This paper proposes the use of factorial experimental design as a standard experimental method in the application of froth flotation to plastic separation instead of the commonly used OVAT method (manipulation of one variable at a time). Furthermore, as is common practice in minerals flotation, the parameters of the kinetic model were used as process responses rather than the recovery of plastics in the separation products. To explain and illustrate the proposed methodology, a set of 32 experimental tests was performed using mixtures of two polymers with approximately the same density, PVC and PS (with mineral charges), with particle size ranging from 2 to 4 mm...

The analysis of genome-wide epigenomic alterations including DNA methylation has become a subject of intensive research for many complex diseases. Whole-genome bisulfite sequencing (WGBS) using next-generation sequencing technologies can be considered the gold standard for a comprehensive and quantitative analysis of cytosine methylation throughout the genome. Several approaches including tagmentation- and post bisulfite adaptor tagging (PBAT)-based WGBS have been devised. Here, we provide a detailed protocol based on a commercial kit for the preparation of libraries for WGBS from limited amounts of input DNA (50-100 ng) using the classical approach of WGBS by ligation of methylated adaptors to the fragmented DNA prior to bisulfite conversion...

Characterization of OFPs (OVATE Family Proteins) has revealed that they exert functions through interacting with different types of transcription factors. However, the molecular bases of these processes are poorly understood. Here, we report that OsOFP19 negatively modulates brassinosteroid (BR) response and integrates it with cell division pattern to affect plant architecture, including grain shape, through interaction with both DLT (DWARF AND LOW-TILLERING) and OSH1 (Oryza sativa homeobox1). Overexpression of OsOFP19 caused semi-dwarf stature with thicker leaves and stronger culms and roots, which is resulted from the increased cell layers in the sub-epidermal tissue...

We report on a 54-year-old man who presented with an atypical, proximal, intraoperative endoleak after endovascular aortic repair with an Ovation endograft for a 65-mm-diameter abdominal aortic aneurysm. The endografting was complicated by inadvertent bilateral iliac limb insertion into the right gate without cannulation of the left gate. The endoleak was treated by brachial approach: Through the open left gate, the outflow inferior mesenteric artery was coil embolized and the inflow left gate was closed with an Amplatzer plug...

The ovate mutation has frequently been used to study changes in fruit shape but not fruit quality. A deterioration in fruit quality associated with the ovate mutation was discovered in this study. To elucidate how ovate influences the quality of fruit, we performed a proteomics analysis of the fruits of the ovate mutant (LA3543) and wild-type ("Ailsa Craig", LA2838A) using tandem mass tag analysis. The results indicated that the ovate mutation significantly influences fruit quality in a number of ways, including by reducing the expression of 1-aminocyclopropane-1-carboxylic acid oxidase 3 (ACO3) in ethylene biosynthesis, improving firmness by reducing the amount of pectinesterase and polygalacturonase, reducing sugar accumulation by downregulating the abundance of mannan endo-1,4-β-mannosidase 4, β-galactosidase, and β-amylase, and reducing the malic acid content by downregulating the accumulation of malic enzymes and malate synthase...

Coronaviruses are of major importance for both animal and human health. With the emergence of novel coronaviruses such as SARS and MERS, the need for fast genome characterisation is ever so important. Further, in order to understand the influence of quasispecies of these viruses in relation to biology, techniques for deep-sequence and full-length viral genome analysis are needed. In the present study, we compared the efficiency of two sequence-independent approaches [sequence-independent single primer amplification (SISPA) and single primer isothermal amplification (SPIA, represented by the Ovation kit)] coupled with high-throughput sequencing to generate the full-length genome of bovine coronavirus (BCoV) from a nasal swab...

OVATE family proteins (OFPs) regulate multiple aspects of plant growth and development; however, their roles in responses to abiotic stresses have not yet been studied. Here, we report that rice OsOFP6 regulates growth and development, and alters responses to drought and cold stresses. OsOFP6 is expressed in various tissues and developmental stages. Knock-down of OsOFP6 by RNA interference (RNAi) resulted in semi-dwarf stature, altered grain shape, and shorter lateral roots. OsOFP6 was shown to interfere with auxin in regulating lateral root growth and initiation, as RNAi plants had increased density of lateral roots under IAA (indole-3-acetic acid) treatment...

Brassinosteroids (BRs) regulate important agronomic traits in rice, including plant height, leaf angle, and grain size. However, the underlying mechanisms remain not fully understood. We previously showed that GSK2, the central negative regulator of BR signaling, targets DLT, the GRAS family protein, to regulate BR responses. Here, we identified Ovate Family Protein 1 (OFP1) as a DLT interacting protein. OFP1 was ubiquitously expressed and the protein was localized in both cytoplasm and nucleus. Overexpression of OFP1 led to enlarged leaf angles, reduced plant height, and altered grain shape, largely resembled DLT overexpression plants...
