Highly effective cystic fibrosis (CF) transmembrane conductance regulator (CFTR) modulator therapies (HEMT), including elexacaftor-tezacaftor-ivacaftor, correct the underlying molecular defect causing CF. HEMT decreases general symptom burden by improving clinical metrics and quality of life for most people with CF (PwCF) with eligible CFTR variants. This has resulted in more pregnancies in women living with CF. All HEMT are known to be able pass through the placenta and into breast milk in mothers who continue on this therapy while pregnant and breast feeding...

Lanosterol, an oxysterol molecule, has been proposed to help maintain lens transparency by inhibiting the formation of protein aggregates. This sterol is produced by the enzyme lanosterol synthase and is part of a metabolic pathway that forms cholesterol as a final step. Abnormalities in lanosterol synthase are responsible for congenital cataracts. The αA-crystallin protein, which acts as a molecular chaperone to lanosterol synthase, has been reported to have anti-protein aggregation, anti-inflammatory and anti-apoptotic properties...

BACKGROUND: Fibrin glue is an absorbable blood-derived product, a biological tissue adhesive which imitates the final stages of the coagulation cascade, it produces a firm clot, forming a seal along the whole length of the wound, the resultant fibrin clot degrades physiologically into granulation tissue 2 weeks after the application. Biological glue has been used extensively in many forms of surgical procedures. Its use in eye surgery has grown lately as we have evidence that showed it was effective in securing conjunctival grafts in pterygium surgery, in securing wounds after glaucoma surgery and more importantly in 20G and 23G vitrectomy...

Chronic diseases (CD) represent the main cause of mortality in developed countries. The increase in the prevalence of of CD is associated with changes in lifestyle habits, including those related to the consumption of processed foodstuffs. In these foods advanced glycation end products (AGE) and advanced lipoperoxydation products (ALE) are formed as a consequence of the reactivity of proteins, carbohydrates, lipid and other components. The aim of the present review is to offer a perspective of how AGE and ALE affect the physiology and development of CD...

We investigated the effects of long-term treatment with probucol, a hypolipidemic agent with antioxidative action, insulin, or their combination on cataracts of streptozotocin-induced diabetic rats fed a high cholesterol diet. Each rat was checked for cataracts at 0, 1, 2, 4, 8, 12 and 15 weeks after streptozotocin injection. Cataracts were observed from 8 weeks in untreated hypercholesterolemic and diabetic rats and the incidence of catarats increased to 100% by 15 weeks. The incidence of cataracts in rats treated with probucol, insulin and their combination was first seen at 12, 12 and 15 weeks, respectively, and was 86%, 63% and 33%, respectively, at 15 weeks...

Serum kynurenine, neopterin, selenium concentrations; neopterin concentrations in aqueous humour from anterior chamber of eye and selenium content in lenses were determined in 40 catarat patients with and in 51--without pseudoexfoliation (PES). Significantly increased kynurenine and neopterin levels in serum and neopterin concentration in aqueous humour were observed in mature cataract patients with PES compared to whose without. These patients also presented with the lowest content of selenium in serum and lens, compared with cataract patients without PES...

In the present study we investigated the effects of a long-term treatment with vitamin E, an antioxidant vitamin, insulin or their combination on cataracts of streptozotocin (STZ)-induced diabetic rats fed a high cholesterol diet. Each rat was checked for cataracts at 0, 1, 2, 4, 8, 12 and 15 weeks after STZ injection. Cataracts were observed from 8 weeks in the control diabetic rats and their incidence of catarats increased to 100% by 12 weeks. The incidence of cataracts in rats treated with vitamin E, insulin and their combination was first seen at 12 weeks and 56%, 20% and 10%, respectively, at 12 weeks and 78%, 50% and 20%, respectively, at 15 weeks...

OBJECTIVE: To study the ultrastructural changes of lenses aged 50 days from normal rats and rats with three kinds of cataract. METHOD: Transmission electron microscope (TEM) was used to investigate the ultrastructural changes of normal lens, selenium, galactosemic and congenital cataracts in the rat. RESULTS: In the normal group, the structure of cell membrane in cortex is not changed obviously, but in the lens nucleus there are slight cellular degeneration, low cytoplasmic density and the destruction of intercellular junctional complexes...

BACKGROUND: The tryptophan degradation pathway leads to NAD production via 3-hydroxykynurenine. Kynurenine aminotransferase (KAT) transforms 3-hydroxykynurenine into xanthurenic acid. In this study, we measured the activity of KAT in human lenses and studied the consequences of xanthurenic acid formation METHODS: KAT activity was determined by the method of Tobes. Fluorescence spectroscopy and SDS-PAGE were used for the protein studies. Thin-layer chromatography and infrared and fast atom bombardment spectrometry were used for substance characterization...

The retinal function may be tested by laser-interference-fringes formed on the retina independently of the refraction status of the eye. The perception of these fringes is a subjective test for the retinal function even in case of moderate opacities of the optical media. Experiences with a new apparatus described here showed that the determination of this "retinal acuity" is very simple to perform. The comparison of retinal acuity and visula acuity of several hundred cataratous eyes and the measurement before and after cataract extraction in about 100 cases are discussed...

Results of combined keratoplasty, extracapsular cataract extraction and lens implantation (triple procedure) are reported for 67 eyes having at least 12 months follow-up. The average age of the patients was 73; the majority were female with a pre-op diagnosis of Fuchs's dystrophy. Post-op visual acuities were 20/40 or better in 72% of the eyes. No implants were removed and only one eye required regrafting. The discission rate was 32%; discissions were usually performed a year post-op. We feel the favorable results are due to uniform use of microsurgical extracapsular catarat extraction at the time of surgery with good capsular fixation of the lens implant...
