RANDOMIZED CONTROLLED TRIAL
A Melka, R Tekle-Haimanot, F Lambien
The efficacy and safety of oral Tolperisone HCL was evaluated in double blind, placebo-controlled, randomized trial in 72 patients with neurolathyrism in stages I, II, and III of the disease at Kolla Duba Health Centre of Dembia District of North Gondar between January and April 1995. Taken orally daily for 12 weeks, tolperisone HCL (Mydocalm) in a dose of 150 milligrams (mgs) twice daily significantly improved subjective complaints such as muscle cramps, heaviness of the legs, startle attacks, flexor spasms and repeated falls...

Mydocalm and nootropil combination was used for treatment of antibiotic neurosensory hypoacusia (24 patients) and non-antibiotic hypoacusis (11 patients). 100 mg of mydocalm was diluted in 10.0 ml of 0.9% sodium chloride and injected intravenously. 20% solution of nootropil (5.0 ml) was injected intramuscularly. The 14-day course produced subjective response. Noise in the ears and heavy head feeling disappeared. As shown by measurements of auditory thresholds, improvement of hearing was only subjective. The responses were similar in both groups of patients...

Photosensitizing and photoprotecting efficiency of about a hundred of compounds, mainly drugs, was studied. The method based on chemiluminescence occurred along with photooxidation of glycyltryptophan under irradiation in UVB range in solution was used for testing. As a measure of photosensitizing efficiency the concentration of photosensitizer which induced two-fold increase of chemiluminescence intensity was chosen. The most effective photosensitizers are riboflavin, FAD, furagin, psoralene, vicasol, benzobarbital, mydocalm, angelicyn, furadonin, ethacridin, diazolin, folic acid...

The efficacy and safety of oral tolperisone hydrochloride (Mydocalm) in the treatment of painful reflex muscle spasm was assessed in a prospective, randomized, double-blind, placebo-controlled trial. A total of 138 patients, aged between 20 and 75 years, with painful reflex muscle spasm associated with diseases of the spinal column or proximal joints were enrolled in eight rehabilitation centers. Patients were randomized to receive either 300 mg tolperisone hydrochloride or placebo for a period of 21 days. Both treatment groups recovered during the 3 weeks rehabilitation program...

A M Kiss, J Martos
The effect of coated Mydocalm tablet, given in 3 x 150-mg daily doses for 3 weeks to 47 patients, simultaneously with physiotherapy, has been examined in comparison to the results of 47 patients treated with physiotherapy solely. In the group of patients receiving Mydocalm as an adjuvant to physiotherapy the alleviation of pain and moderation of muscular hypertonia and spasm were observed within a shorter period. On the basis of the results of these examinations the use of Mydocalm is recommended as an adjuvant to complex therapy, in 300-450-mg daily doses, for the treatment of locomotor diseases accompanied by muscular hypertonia, muscular spasticity, and contracture...

The antagonistic effect of 2,4'-dimethyl-3-piperidino propiophenone hydrochloride (Mydocalm) and its 15 derivatives against the activity of slow reacting substance of anaphylaxis (SRS-A) were examined in vitro. Mydocalm, 2-methyl-3-piperidino-beta-propionaphthone hydrochloride (As-5) and 2,3',4'-trimethyl-3 piperidinopropiophenone hydrochloride (As-14) were found to be potent antagonists to SRS-A. The above three compounds inhibited homologous passive cutaneous anaphylaxis (PCA) in rats and guinea pigs but not heterologous PCA in guinea pigs...
