The elevated expression of NQO1 in many human solid tumors along with its ability to activate quinone-based anticancer agents makes it an excellent target for enzyme-directed drug development. NQO1 plays an important role in melanogenesis and given its correlation with a poor patient outcome we propose this enzyme as an intriguing target for molecular-based therapeutic regimen against melanoma. Chemotherapy, the most commonly used therapeutic method for cancer, has the inherent constraint of low bioavailability. A number of physical cancer therapeutic treatments like radiation, ultrasound, photo-acoustic/photo thermal, microwave therapies are based on locating the afflicted sites with the help of imaging, but the serious drawbacks of these treatment options are that they damage the neighboring normal tissues and/or induce undesired cancer metastasis. Herein, a novel drug photorelease system based on gold nanostars (AuNSts), coated with a mesoporous silica shell and capped with paraffin as thermosensitive molecular gate, is reported. Direct measurements of the surface temperature of a single gold nanostar irradiated using a tightly focused laser beam are performed via a heat-sensitive biological matrix.
