Deubiquitylating enzymes (DUBs) play a vital role in the ubiquitin pathway by editing or removing ubiquitin from their substrate. As breakthroughs within the ubiquitin field continue to highlight the potential of deubiquitylating enzymes as drug targets, there is increasing demand for versatile high-throughput (HT) tools for the identification of potent and selective DUB modulators. Here we present the HT adaptation of the previously published MALDI-TOF-based DUB assay method. In a MALDI-TOF DUB assay, we quantitate the amount of mono-ubiquitin generated by the in vitro cleavage of ubiquitin chains by DUBs...

Nanoindentation technology with high spatial resolution and force sensitivity is widely used to measure the mechanical properties of hard biomaterials and tissues. However, its reliability to analyze soft biomaterials and organs has not been tested. Here, we evaluated the utility of nanoindentation to measure the passive mechanical properties of soft biological specimen. Kidney, liver, spleen and uterus samples were harvested from C57BL/6 N mice. We assessed test-retest repeatability in biological specimen and hydrogel controls using Bland-Altman diagrams, intraclass correlation coefficients (ICCs) and the within-subject coefficients of variation (COVs)...

The multi-billion dollar trade in ornamental fishes has rarely been reliably monitored. Almost all coral reef fishes are wild-caught, and few scientific analyses have attempted to elicit exact quantities and identify species involved. The consequences of the removal of millions of these fishes are poorly understood. This article collates and examines available information, including scientific studies and formal publications, in order to create a more accurate picture of this commerce. We demonstrate that it is almost impossible to analyse the trade in marine ornamental fishes due to a lack of data, and that available data for marine species is frequently combined with that for freshwater species...

PURPOSE: To translate and culturally adapt the Patient and Observer Scar Assessment Scale, POSAS, to Norwegian and explore its test-retest, intra- and inter-tester reliability. METHODS: POSAS was translated into Norwegian following international guidelines in collaboration with an international translation bureau. Twenty-six adults and 24 children were recruited from a burns outpatient clinic. Three observer-categories: doctor, nurse and physiotherapist, assessed the patients' scars and scored the Observer scale for estimating inter-tester reliability...

Ionic liquids (ILs) as green solvents have been studied in the application of gas sweetening. However, it is a huge challenge to obtain all the experimental values because of the high costs and generated chemical wastes. This study pioneered a quantitative structure-property relationship (QSPR) model for estimating Henry's law constant (HLC) of H2 S in ILs. A dataset consisting of the HLC data of H2 S for 22 ILs within a wide range of temperature (298.15-363.15 K) were collected from published reports. The electrostatic potential surface area (SEP ) and molecular volume of these ILs were calculated and used as input descriptors together with temperature...

BACKGROUND: The ESCALOX trial was designed as a multicenter, randomized prospective dose escalation study for head and neck cancer. Therefore, feasibility of treatment planning via different treatment planning systems (TPS) and radiotherapy (RT) techniques is essential. We hypothesized the comparability of dose distributions for simultaneous integrated boost (SIB) volumes respecting the constraints by different TPS and RT techniques. The global spread of the novel coronavirus (SARS-CoV-2) and increasing rate of mortality among different countries has raised the global concern regarding this disease. This illness is able to infect human beings through person-to-person contact at an extremely high rate. World Health Organization proclaimed that COVID-19 disease is known as the sixth public health emergency of international concern (30 January 2020) and also as one pandemic (12 March 2020). Owing to the rapid outbreak of COVID-19 worldwide, health authorities focused on discovery of effective prevention and treatment techniques for this novel virus...

High pressure-temperature conditions can be readily achieved through the laser-heated diamond anvil cell (LH-DAC). A stable laser source is required for reliable in situ measurements of the sample, as the sample is small with a thermal time constant of the order of microseconds. Here, we show that the power instabilities typical of CO2 gas lasers used in LH-DAC's are ±5% at the second timescale and ∼±50% at the microsecond timescale. We also demonstrate that the pointing instability of the laser requires either a diffuser or an integrating sphere for reliable total power measurements with small sized detectors...

Development of lithium ion batteries with ultrafast charging rate as well as high energy/power densities and long cycle-life is one of the imperative works in the field of batteries. To achieve this goal, it requires not only to develop new electrode materials but also to develop nano-characterization techniques that are capable of investigating the dynamic evolution of the surface/interface morphology and property of fast charging electrode materials during battery operation. Although electrochemical atomic force microscopy (EC-AFM) holds high spatial resolution, its imaging speed is too slow to visualize dynamics occurring on the timescale of minutes...

We report a straightforward beam splitter in the soft x-ray spectral range using a thin oxidized aluminum foil. As it allows us to monitor reliably shot-to-shot variations in energy and in energy distribution, this beam splitter is of high interest for the simultaneous use of diagnostics for soft x-rays sources. We measure a transmission of 0.5 and a reflectivity of 0.018 at 22.5° of incidence with a soft x-ray laser working at 32.8 nm. These values are in good agreement with the theory. As the theory predicts a reflectivity and a transmission of both 12% at 52...

We present the Aurore platform for ultrafast sciences. This platform is based on a unique 20 W, 1 kHz, 26 fs Ti:sapphire laser system designed for reliable operation and high intensity temporal contrast. The specific design ensures the high stability in terms of pulse duration, energy, and beam pointing necessary for extended experimental campaigns. The laser supplies 5 different beamlines, all dedicated to a specific field: attosecond science (Aurore 1), ultrafast phase transitions in solids (Aurore 2 and 3), ultrafast luminescence in solids (Aurore 4), and femtochemistry (Aurore 5)...

The optimized extraction was run on a BioSprint 15 platform. BACKGROUND: Exposure to organophosphate flame retardants and plasticizers (PFRs) is commonly estimated by measuring biomarker concentrations in spot urine samples. However, their concentrations in urine can vary greatly over time due to short biological half-lives and variable exposure, potentially leading to exposure misclassification.
