Chemotherapy-induced peripheral neuropathy (CIPN) is a common side effect of cancer treatment. No medication has been shown to be effective in the treatment of CIPN. This study aims to integrate the image-based high-content screening, mouse behavior models and mechanistic cell-based assays to discover potential neuroprotective drugs. In this study, we evaluated the anti-alopecia and anticancer effects of PAK1 inhibitors isolated from Alpinia zerumbet (alpinia) in cell culture. Free intracellular calcium ([Ca(2+)]i), in addition to being an important second messenger, is a key regulator of many cellular processes including cell membrane potential, proliferation, and apoptosis. In many cases, the mobilization of [Ca(2+)]i is controlled by intracellular store activation and calcium influx. We have investigated the effect of several ion channel modulators, which have been used to treat a range of human diseases, on [Ca(2+)]i release, by ratiometric calcium imaging. Minoxidil is clinically used to prevent hair loss. However, its effect on Ca(2+) homeostasis in prostate cancer cells is unclear. This study explored the effect of minoxidil on cytosolic-free Ca(2+) levels ([Ca(2+)]i) and cell viability in PC3 human prostate cancer cells.
