The northern elephant seal, Mirounga angustirostris, was heavily hunted and declared extinct in the 19th century. Basic fibroblast growth factor (bFGF) can stimulate wound healing. However, consistent delivery of bFGF has many disadvantages. To decrease their instability and diffusible, we introduced chitin-binding domain (ChtBD) into bFGF. Two expression plasimids were constructed. The first one (named bFGF) contained bFGF (154 amino acids), the second (named ChtBD-bFGF) contained bFGF and the ChtBD (54 amino acids). The present contribution aims at describing the fabrication of coacervates in the nano-size range starting from a 1-deoxylactit-1-yl chitosan (in this manuscript termed as CTL60) and the multivalent anion tripolyphosphate (TPP). Colloidal coacervates have been obtained for precise values of the molar ratio of TPP to CTL60 repeating unit. Coacervation is ensured only at pH 4.5 and not at 7.4, thus demonstrating the key role of electrostatic interactions in the stabilization of the coacervates. At a variance with chitosan, CTL60 favors the formation of highly homogeneous coacervates with very low values of the polydispersity index (PDI)...

Salecan is a novel water-soluble extracellular β-glucan and has excellent physicochemical and biological properties. A series of hydrogels were fabricated from Salecan and poly(2-acrylamido-2-methylpropanesulfonic acid-co-N-hydroxymethyl acrylamide) (poly(AMPS-co-HMAA), PAH) by semi-IPN technique. The introduction of Salecan into the PAH network endows the system with enhanced cell adhesion properties. Chitooligosaccharide (COS) has been shown to regulate many biological functions, such as antimicrobial effect and antitumor activity. In the present study, highly N-acetylated chitooligosaccharide (NACOS) was prepared by N-acetylation of COS, and the anti-inflammatory activity of NACOS in macrophages were evaluated. The results indicated NACOS significantly suppressed the LPS-induced pro-inflammatory cytokines interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) expression. A simple, fast and accurate spectrophotometric method had been developed to determine lead (II), chromium (III) and barium (II) ions in pure forms and in spiked water samples using thoron (THO) as a reagent forming colored complexes. It was found that the formed complexes absorbed maximally at 539, 540 and 538nm for Pb(II)-THO, Cr(III)-THO and Ba(II)-THO complexes, respectively. The optimum experimental conditions for these complexes had been studied carefully. Organic fluorophores have a wide range of biological uses and are usually needed to be prepared as water-soluble compounds or nanoparticles for applications in aqueous biosystems owing to their hydrophobic properties, which often is a complex, time-consuming and high-cost process. Here, the nanoparticle preparation of hydrophobic fluorophores and their application in cell imaging have been investigated. In this issue of Cell Chemical Biology, Pezacki and co-workers (Sherratt et al., 2017) report a simple method to metabolically label viable bacteria, which can be used to detect and capture foodborne pathogens. The method may also find many other applications because it can be used to recover live cells, pathogens and non-pathogens, from various biomedical and environmental samples.

In order to develop a streptavidin-biotin system with optimal performance in fluorescent labeling, in this issue of Cell Chemical Biology, Jacobsen et al. (2017) focused on changing the surface density of amino groups present on streptavidin via lysine mutagenesis. The streptavidin mutant containing only one free amino group was found superior to other streptavidin variants and was named Flavidin for fluorophore-friendly streptavidin.
