In the course of our validation program testing blood-brain barrier (BBB) in vitro models for their usability as tools in drug discovery it was evaluated whether an established Transwell model based on porcine cell line PBMEC/C1-2 was able to differentiate between the transport properties of first and second generation antihistaminic drugs. First generation antihistamines can permeate the BBB and act in the central nervous system (CNS), whereas entry to the CNS of second generation antihistamines is restricted by efflux pumps such as P-glycoprotein (P-gP) located in brain endothelial cells...

Cetirizine (Zyrtec) is a potent and long-acting second-generation histamine H1- receptor antagonist for the treatment of allergic disease, such as allergic rhinitis and chronic idiopathic urticaria, in adult and child. It is a racemic mixture of levocetirizine (Xyzal) and dextrocetirizine. The purpose of this present study was to compare the efficacy of cetirizine, levocetirizine and placebo for the treatment of pediatric perennial allergic rhinitis. 74 perennial allergic rhinitis patients, aged 6 to 12 years old, assigned to 1 of 3 treatment groups for 12 weeks randomly...

A simple, rapid and sensitive spectrophotometric method has been developed for the assay of ceterizine hydrochloride (CTZH) in bulk drug and its pharmaceutical preparations. This method is based on the ion-pair complex reaction between CTZH and Alizarin Red S in Clarks-Lubs buffer. The chromogen being extractable with chloroform, could be measured quantitatively at 440 nm. All variables were studied to optimise the reaction conditions. Regression analysis of Beer's Law plot showed good correlation in the concentration range 2...

An accurate, simple, reproducible and sensitive HPLC method for the determination of ceterizine hydrochloride (CTZH) and paracetamol (PARA) has been developed and validated. The separation of CTZH, PARA and Nimesulide (the internal standard) was achieved on a CLC C(18) (5 mu, 25 cm x 4.6 mm i.d) column using UV detection at 230 nm. The mobile phase was consisted of acetonitrile-water (55:45 v/v). The linear ranges of detection for CTZH and PARA were found to be 0.715-55 microg/ml (r(2)=0.9985) and 0.55-39 microg/ml (r(2)=0...

Two simple, rapid and sensitive extractive spectrophotometric methods have been developed for the assay of ceterizine hydrochloride (CTZH) in bulk drug and in pharmaceutical preparations. These methods are based on the formation of chloroform soluble complexes between CTZH with bromocresol purple (BCP) or bromophenol blue (BPB) in Walpole buffer of pH 2.64 with an absorption maximum at 409 nm and at 414 nm for BCP and BPB, respectively. Reaction conditions were optimised to obtain the maximum colour intensity...

To investigate the role of trigeminal subnucleus caudalis in neural mechanisms of irritation, we recorded single-unit responses to application of a variety of irritant chemicals to the tongue or ocular mucosa in thiopental-anesthetized rats. Recordings were made from wide dynamic range (WDR) and nociceptive-specific units in superficial layers of the dorsomedial caudalis (0-3 mm caudal to obex) responsive to mechanical stimulation and noxious heating of the ipsilateral tongue ("tongue" units) and from WDR units in ventrolateral caudalis (0-2 caudal to obex) responsive to mechanical and noxious thermal stimulation of cornea-conjunctiva and frequently also surrounding skin ("cornea-conjunctival" units)...

An effect of cetirizine--one of antihistaminic agents--on the rate of adverse reactions to the specific desensitization has been evaluated in patients with atopic respiratory diseases. The study included 270 patients desensitized in 1988-1993 for the atopic diseases due to hypersensitivity to grass pollens and Dermatophagoides pteronyssinus. Standard Alvac-P. Pollinex, and Alvac-S HDM vaccines have been used. Occurrence and severity of the adverse reactions noted during 3,368 injections of vaccines have been analysed...

A method using reversed-phase high-performance liquid chromatography with ultraviolet detection for the determination of ceterizine in serum is described. The method is sensitive down to 50 ng/ml (250-microliter loop). Sample preparation involves only serum deproteination with perchloric acid and injection of the centrifuged supernatant. Elution is at pH 2.5 with acetonitrile-methanol-0.05 M phosphate buffer (33:9:58, v/v) on a 25 cm x 4.6 mm I.D. Spherisorb S5 ODS2 column. Detection is at 211 nm, its lambda max...
