Amphibians are experiencing dramatic worldwide declines and many species are reliant on captive breeding programs to ensure continued survival. However, captive breeding in amphibians is not always successful because many species, especially ones in decline, have particular and specific breeding needs. The endangered alpine tree frog, Litoria verreauxii alpina , has never been bred in captivity before. Due to its dramatic declines across the Australian Alps caused by the global pandemic chytridiomycosis, the species is a potential candidate for captive assurance colonies, which rely on captive breeding...

CONTEXT: With global amphibian biodiversity rapidly declining, improving reproductive technology outcomes has become essential. Captive breeding programs have struggled because amphibian breeding physiology often requires specific environmental cues that reproductive technologies can circumvent. AIMS: This study tests the efficiency of hormonal induction by evaluating sperm quality in the endangered Litoria verreauxii alpina . METHODS: We assessed the effects of exogenous hormones - gonadotrophin-releasing hormone agonist (GnRH-a, Lucrin), and human chorionic gonadotrophin (hCG, Chorulon) - on sperm quality...

The study was conducted to compare estrous rate, ovulatory response, plasma progesterone concentrations, and conception rate following cervical artificial insemination in goats given a new or once-used controlled internal drug release (CIDR) device with human chorionic gonadotropin (hCG). Fifty-six Thai-native goats with the average age and body weight of 11 months and 17.3 kg received a 14-day treatment with a new CIDR device (Eazi-Breed(TM)CIDR®, Pfizer, NY, USA) or a once-used CIDR device. All goats received a 300-IU injection of hCG (Chorulon®, Intervet International B...

Reproductive management programs that synchronize ovulation can ovulate a smaller than normal follicle, potentially resulting in inadequate progesterone (P4) concentrations after artificial insemination (AI). Ovulation of the dominant follicle of the first follicular wave with human chorionic gonadotropin (hCG) treatment can produce an accessory corpus luteum and increase circulating P4 concentrations. This manuscript reports the results of 2 separate analyses that evaluated the effect of hCG treatment post-AI on fertility in lactating dairy cows...

Our objectives were to (1) compare the effect on pregnancies per artificial insemination (P/AI) of presynchronization of the estrous cycle with human chorionic gonadotropin (hCG) 7d before resynchronization of ovulation (Resynch) initiated 25 d after timed artificial insemination (TAI) and compare the presynchronization treatment with the Double-Ovsynch (DO) protocol, and (2) evaluate whether hCG for presynchronization could be replaced with GnRH. In experiment 1, lactating Holstein cows were blocked by parity and were randomly assigned to receive (1) Resynch-25 (D25), the Resynch protocol (GnRH-7 d-PGF(2 α)-56 h-GnRH-16 h-TAI) initiated 25 d after TAI (n=418); (2) HGPG, presynchronization with hCG (2,000 IU of Chorulon) 7d before D25 (n=450); and (3) DO (Pre-Resynch, GnRH-7 d-PGF(2 α)-72 h-GnRH; Breeding-Resynch, GnRH-7 d-PGF(2 α)-56 h-GnRH-16 h-TAI) initiated 22d after TAI (n=405)...

The present investigation addresses the pharmacokinetics of human chorionic gonadotropin (hCG), intramuscularly (i.m.) administered to goats. Nine pluriparous does of the Boer goat breed, 2-6 years of age and weighing 45-60 kg, were administered 500 IU hCG (2 ml Chorulon) deep into the thigh musculature 18 h after superovulatory FSH treatment. Blood samples were drawn from the jugular vein at 2  h intervals for the first 24h, at 6 h intervals until 42 h, and at 12 h intervals until 114 h after administration...

The purpose of the present study was to hasten the resumption of ovarian activity early postpartum in lactating dairy cows, using equine chorionic gonadotropin (eCG), to enhance follicular growth, followed by hCG, to induce ovulation. Primiparous Holstein dairy cows (n=21) were assigned equally into eCG, eCG-hCG and Control groups. Cows in the eCG and eCG-hCG groups received an i.m. injection of eCG (500 IU Folligon®) on Day 6 postpartum. Cows in the eCG-hCG group were also given an i.m. injection of hCG (500 IU Chorulon®), once dominant follicle reached the diameter of 13-16 mm following eCG injection...

The aim of this investigation was to optimize fixed-time insemination in goats by clustering ovulations in prostaglandin F(2α)-synchronized goats either with gonadotropin releasing hormone (GnRH) or human chorionic gonadotropin (hCG). The underlying intention was to reduce the incidence of short cycles by providing a more sustained stimulation of the corpus luteum by substituting the commonly used GnRH with longer-acting hCG. It was conjectured that this might render the corpus luteum less prone to premature regression...

The spotted green pufferfish (Tetraodon nigroviridis) is an important genetics model animal due to its small, well-mapped genome. However, only wild-caught juveniles and adults are available to researchers. A lack of gametes, fertilized eggs, developing embryos, and other early life stages hampers development of the full potential of T. nigroviridis as a model research species. We report on successful spawning trials using a novel induced spawning technique, ovarian lavage. Chorulon(®) (human chorionic gonadotropin, hCG) was injected into a catheter inserted into the oviduct at a rate of 3 μl/g body weight...

We determined the effects of administering hCG 7 d after a fixed-time AI (TAI) on ovarian response, concentrations of progesterone, and pregnancy rates in postpartum suckled beef cows. Cows at 6 locations received 100 microg of GnRH (Fertagyl, Intervet Animal Health, Millsboro, DE) and a controlled internal drug release (CIDR) device (CIDR EAZI-Breed, Pfizer Animal Health, New York, NY), followed in 7 d by 25 mg of PGF(2alpha) (Lutalyse, Pfizer Animal Health) and CIDR removal. At 64 h after CIDR removal, cows received an injection of GnRH and AI (d 0), and then were stratified by days postpartum and parity and assigned randomly to 2 treatments administered 7 d after TAI: 1) 1 mL of saline (saline; n = 252); or 2) 1,000 IU of hCG (Chorulon, Intervet Animal Health; n = 254)...

The objectives of this study were to determine the effect of a single intramuscular injection of 200IU hCG (Chorulon) on Day 12 post-mating on ovarian function and subsequent lambing performance in ewes and ewe lambs bred at synchronised oestrus during the breeding season and on the lambing performance of ewes induced to breed during late anoestrus. All animals were mated to rams at synchronised oestrus and on Day 12 post-mating given normal saline or 200IU hCG. An efficient protocol was developed to synchronize and superovulate mature pigs for the collection of pronuclear stage embryos suitable for DNA microinjection. A timed and coordinated regimen of Lutalyse, PG600 and Chorulon along with daily checking for estrus allowed synchronization of groups of gilts having estrous cycles at regular intervals. Pigs 10-16 days after the beginning of standing estrus have been successfully synchronized into estrus using this protocol. REASONS FOR PERFORMING STUDY: To compensate for the wide variation in the freezability of stallion spermatozoa, it has become common veterinary practice to carry out repeated ultrasonography of the ovaries of oestrous mares in order to be able to inseminate them within 6-12 h of ovulation with a minimum of 300-500 x 10(6) frozen-thawed spermatozoa. Furthermore, in order to achieve satisfactory fertility, this requirement for relatively high numbers of spermatozoa currently limits our ability to exploit recently available artificial breeding technologies, such as sex-sorted semen, for which only 5-20 x 10(6) spermatozoa are available for insemination...
