Reactive oxygen species (ROS) are involved in the mechanism of photoaging and carcinogenesis. Skin is endowed with antioxidant enzymes including superoxide dismutases (SOD): cytosolic copper zinc SOD and mitochondrial manganese SOD. The aim of our study was to estimate the protective effect of manganese against oxidative injury on cultured human skin fibroblasts. Dithranol, hydrogen peroxide and UV-A radiation (375 nm) were employed as oxidative stressors. The supply of manganese chloride produced an increase in cellular content of this element up to 24 fold without concomitant elevation of MnSOD activity...

The authors propose a new immunoenzymatic method (ELISA) for human brucellosis diagnosis. They study in a logical way some of the parameters of the reaction realizable by macro- or microassay. The all method requires less than six hours. They use OMS recommended brucella suspension as antigen. The conjugate enzyme is peroxydase and human anti-immunoglobulins are either complete or specific IgM. The revelating substrate is obtained by association of hydrogen peroxyde with 5 amino-salicylic acid which gives the best colorimetric stability...

A new solid phase enzyme-linked immunosorbent assay (ELISA) was developed for the quantitation of human protein C antigen. Anti-protein C F(ab')2 fragments were adsorbed to polystyrene plates. The binding of serial dilutions of control or test plasma, containing protein C, was detected by incubation with peroxidase-labeled anti-protein. C-IgG followed by the addition of hydrogen peroxyde and 0-phenylenediamine. This ELISA is specific, sensitive (detection limit: 0.02%) and accurate (variation coefficient: 3 to 10%)...

A new solid phase enzyme-linked immunosorbent assay (ELISA) was developed for the quantitation of human Factor VII antigen (F VII Ag), using a monospecific rabbit anti-F VII antiserum. Anti-F VII F(ab')2 fragments were adsorbed to polystyrene plates. The binding of serial dilutions of control or test plasma, containing F VII, was detected by incubation with peroxidase-labeled anti- FV II IgG followed by the addition of hydrogen peroxyde and O-phenylenediamine. This ELISA is specific, sensitive (detection limit: 0...

Superoxide radicals produced during acute intestinal ischemia are biochemically related with the presence of hydrogen peroxyde. In this study we have investigated the distribution of peroxidase-catalase activity, histochemically determined, in the ischemic ileal wall. In the rat, complete arterial and venous occlusion produced a progressive increase in extra-vascular peroxidase-catalase activity with a maximum corresponding to the ileal wall. Probably the tissue peroxidase-catalase activity is related to massive degranulation of polymorphonucleates...

A fluorimetric, authomatical method for determination of free and total cholesterol in serum is described. Cholesterol esters are splitted into free cholesterol and fatty acids by cholesterol esterase. In the presence of oxygen free cholesterol will be transformed by cholesterol oxydase in delta 4 cholestenone with formation of hydrogen peroxyde. Hydrogen peroxyde oxidizes in the presence of catalase inethanol to formaldehyde which gives with ammonium ions and acetylacetone 3-5 diacetyl 1-4 dihydrolutidine measured by fluorimetry...

High affinity estradiol binding is found in the fraction of human eosinophil rich, polynuclear leukocytes sedimenting between 800 times 10 g times mn and 24,000 times 20 g times mn. The apparent dissociation constant at equilibrium is approximately 0.6 nM, and the total number of binding sites per blood eosinophil leukocyte is 7,400 sites per cell. Hydrogen peroxyde (1 muM) enhances estradiol binding, a finding similar to that previously observed autoradiographically for the binding of estradiol to the eosinophils of rat uterus...
