The metabolism of flavonoids derived from orange juice in Chinese volunteers has not been well investigated. With the ultra-fast liquid chromatography-quadrupole-time-of-flight tandem mass spectrometry (UFLC-Q-TOF-MS/MS) system, orange juice-derived flavonoids, as well as metabolites contained in urine collected from healthy Chinese volunteers after consumption of 250mL orange juice, were systematically identified and quantified. Finally, a total of 9 flavonoids and 30 metabolites were detected. We have employed size exclusion chromatography (SEC) to the study of the auto- and Cu(2+)-mediated oxidation of the catecholamines, dopamine, epinephrine and norepinephrine, into melanins. We observed that, due to non-size exclusion-mediated effects, the catecholamines and some of the low molecular mass intermediates generated during the oxidation reactions, could be resolved from each other and from the high molecular mass pigment generated.
