ESKAPE pathogens (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacter species) constitute a threat to humans worldwide. We analyzed the rates of antimicrobial resistance in ESKAPE pathogens across India over the 2010-2020 decade after retrieving the data from the Medline PubMed repository using specific keywords. 89 studies that included 20,177 ESKAPE isolates showed that A. baumannii isolates were the most represented (35.9%, n = 7,238), followed by P...

BACKGROUND: With increasing antibiotic resistance and regulation, the issue of antibiotic combination has been emphasized. However, antibiotic combination prescribing lacks a rapid identification of feasibility, while its risk of drug interactions is unclear. METHODS: We conducted statistical descriptions on 16,101 antibiotic coprescriptions for inpatients with bacterial infections from 2015 to 2023. By integrating the frequency and effectiveness of prescriptions, we formulated recommendations for the feasibility of antibiotic combinations...

Allometric dose scaling aims to create isometric exposures between animals and humans and is often employed in preclinical pharmacokinetic/pharmacodynamic models. Bolus dose allometric scaling is the most simple and commonly used strategy in kidney injury studies; however, it is possible to humanize drug exposures. Currently, it is unknown if dose-matched bolus allometric scaling results in similar outcomes to humanized infusions in the vancomycin induced kidney injury model. We utilized a preclinical Sprague-Dawley rat model to compare traditional allometrically scaled dose-matched bolus administration of vancomycin to a computer-controlled and humanized infusion scheme to assess for differences in iohexol-measured kidney function and urinary kidney injury biomarkers...

OBJECTIVE: Imipenem-relebactam (IMR), a novel β-lactam/β-lactamase inhibitor combination, is recommended for infections caused by difficult-to-treat Pseudomonas aeruginosa. This study aimed to investigate the evolution trajectory of IMR resistance under the selection of levofloxacin in P. aeruginosa. METHODS: Antimicrobial susceptibility testing, complete genome sequencing and gene manipulation experiments were performed. Quantitative reverse transcription PCR for specific genes and porin levels were detected...

OBJECTIVES: This study aimed to appraise clinical practice guidelines (CPGs) for the treatment of carbapenem-resistant Gram-negative bacilli (CRGNB) infections and to summarize the recommendations. METHODS: A systematic search of the literature published from January 2012 to March 2023 was undertaken to identify CPGs related to CRGNB infections treatment. The methodological and reporting quality of eligible CPGs were assessed using six domains of the Appraisal of Guidelines for Research and Evaluation (AGREE) II tool and seven domains of the Reporting Items for practice Guidelines in HealThcare (RIGHT) checklist...

Successful viral propagation within infected cells necessitates the viruses' ability to overcome the cellular integrated stress response (ISR), triggered during viral infection, which in turn inhibits general protein translation. In our study, we unveil a shared tactic employed by viruses to suppress ISR by upregulating host cell polyribonucleotide nucleotidyltransferase 1 (PNPT1). The propagation of adenovirus, murine cytomegalovirus, and hepatovirus within their respective host cells induces PNPT1 expression...

INTRODUCTION: The COVID-19 pandemic has further highlighted the continuing threat of antimicrobial resistance (AMR) to global health and economic development. In the last two decades, AMR has raised increasing concern with an estimated 4.95 million deaths globally due to bacterial AMR in 2019 alone. This study aims to analyze the impact of the pandemic on the spread of multi-drug resistant organisms (MDROs) using data from the Hospital «P. Giaccone» in Palermo, comparing pre-pandemic and pandemic periods...

Human adenovirus (HAdV) and cytomegalovirus (HCMV) are cause of high morbidity and mortality in patients receiving solid organ (SOT) and hematopoietic stem cell transplantations (HSCT). Immunosuppressors are universally used to prevent graft-versus-host disease (GVHD) in HSCT and graft rejection in SOT. The long-term use of these drugs is associated with a high risk of infections, but there are also evidences of their specific interference with virus infection. The antiviral activity of immunosuppressors commonly used in the clinical practice in SOT and HSCT recipients has been evaluated in vitro to determine whether their use could be associated with less risk of HAdV and HCMV infection...

The control and elimination of malaria caused by Plasmodium vivax is hampered by the threat of relapse infection resulting from the activation of dormant hepatic hypnozoites. Currently, only the 8-aminoquinolines, primaquine and tafenoquine, have been approved for the elimination of hypnozoites, though their use is hampered by potential toxicity. Therefore, an alternative radical curative drug that safely eliminates hypnozoites is a pressing need. In this study, we assessed the potential hypnozoiticidal activity of the antibiotic azithromycin, which is thought to exert antimalarial activity by inhibiting prokaryote-like ribosomal translation, within the apicoplast, an indispensable organelle...

Aztreonam-avibactam is under clinical development for treatment of infections caused by carbapenem-resistant Enterobacterales (CRE), especially those resistant to recently approved β-lactamase inhibitor combinations (BLICs). We evaluated a large collection of CRE isolates, including those nonsusceptible to ceftazidime-avibactam, meropenem-vaborbactam, and/or imipenem-relebactam. Overall, 24,580 Enterobacterales isolates were consecutively collected (1/patient) in 2020-2022 from 64 medical centres located in Western Europe (W-EU), Eastern Europe (E-EU), Latin America (LATAM), and the Asia-Pacific region (APAC)...

BACKGROUND: Oseltamivir, as a low-cost antiviral agent, may support or complement treatment of COVID-19. This study assessed whether oseltamivir is effective in reducing COVID-19 related mortality. METHODS: This retrospective cohort study investigated real world data from a nationwide database of hospitalization due to severe acute respiratory syndrome in Brazil. We used propensity score matching to mimic a randomized controlled trial with "oseltamivir" and "no antivirals at all" as the intervention and control groups, respectively...

To understand the global changes in the nonsusceptibility rates of Escherichia coli to meropenem and ceftazidime-avibactam (CZA), we conducted a study using the Antimicrobial Testing Leadership and Surveillance database. A total of 49,394 E. coli isolates were collected during the 8-year study period. The countries with the highest nonsusceptible rates for meropenem were India (16.6%), followed by Pakistan (6.7%), Ukraine (5.4%), Oatar (5.3%), and Guatemala (3.2%). For CZA, the nonsusceptible rate was highest in India (15...
